Build 1×1 rooms, with lockable doors. Put one nest box in each room. Create a 1×1 pasture zone on top of the nest box, and assign one female egg-layer to it.

Up stairs: dig out the block. Down stairs: channel a hole in the floor, or place a new floor tile over top of it. Up/Down combo: simply digging out the tile should remove both parts of the stairs.
